The Effect of Actinomycin S on RNA Synthesis in PS Y15¡¯ Cells

Abstract
The actinomycin is known that it inhibits the DNA-dependent cellular RNA :synthesis (Hurwitz et al, 1962; Goldburg et al, 1962; Levinthal et al, 1963; Reich et al, 1963) and it presumably forms a complex with the cellular DNA (Kawamata and Imanishi, 1960; Rauen et al, 1960; Kirrk, 1960).
The actimomycin S is derived from a somatic and extracellular substances of a certain strain of streptomyces (Nobuo, 1959) and it has three components; S1, S2, and S3(Fujita, 1959).
According to the recent study of paper chromatography, it seems to be that actinomycin S2 has almost the same structure with that of actinomycin D, and L-proline in one peptide chain of actinomycin D is replaced by a gamma-oxo-proline in the actinomycin S3 (Kawamata, 1966), the effect of actinomycin S on DNA might be similar to that of actinomycin D.
In evident, 3H-actinomycin S3 was preferentially accumulated in the region of nucleus in human TG cells (Kawamata, et a], 1965).
In this paper the inhibition of RNA synthesis by actinomycin S was demonstrated in porcine kidney (PS Y15¢¥) cells.
II. Materials and Methods
Cell culture
The cells of porcine kidney stable cell line (PS Y15¢¥), kindly supplied by Dr. Y. K. Inoue, Institute for Virus Research, Kyoto University, have been cultivated in TC-199 medium (Difco. laboratory, USA) supplemented with 5 per cent of inhibitor free calf serum. The monosheets of the cells were obtained by seeding 106 cells in 5 ml of the same medium per prescription bottle ( 2 oz) and incubating for three days.
Inhibited by the increasing doses of actinomycin. Most of cells, which were treated. with AMS 0.5pg/m1 for 2.5 hours, were almost completely inhibited the RNA synthesis for a period in a drug free medium.
